Why Molecular Size Matters
All AHAs work by loosening the bonds between dead skin cells at the surface, accelerating cell turnover. But molecular weight determines penetration rate:
- Glycolic acid: ~76 Da — smallest AHA, fastest penetration, highest irritation potential
- Lactic acid: ~90 Da — moderate penetration, better tolerated
- Mandelic acid: ~152 Da — largest common AHA, slowest penetration, lowest irritation potential
Slower penetration means more even exfoliation, less risk of over-exfoliation in thin or sensitive areas, and significantly reduced post-inflammatory hyperpigmentation (PIH) risk — making mandelic acid particularly well-suited for Fitzpatrick skin types IV–VI.

The Evidence for Acne
Mandelic acid has a dual mechanism for acne: it is both an exfoliant and an antimicrobial. Its lipophilic structure allows it to penetrate sebaceous follicles more effectively than purely hydrophilic AHAs.
Sarkar et al. (2013), Journal of Cosmetic Dermatology, PMID 23438143 — compared mandelic acid peels to salicylic-mandelic combination peels in acne patients with darker skin tones. Both showed significant improvement in acne lesion counts with minimal PIH.
Khunger et al. (2011), Journal of Cutaneous and Aesthetic Surgery, PMID 21976882 — found mandelic acid peels effective for both acne and post-acne hyperpigmentation in Indian skin, with a favorable safety profile.

The Evidence for Hyperpigmentation
Mandelic acid inhibits tyrosinase — the enzyme responsible for melanin synthesis — through a mechanism similar to kojic acid.
Ahn & Kim (2010), Annals of Dermatology, PMID 21165219 — documented tyrosinase inhibition by mandelic acid in vitro, supporting its use as a brightening agent.
Evidence Tier 2: Mechanistic evidence is strong; clinical brightening data is promising but requires more large-scale RCTs.
Who Should Use Mandelic Acid
- Sensitive or reactive skin — including rosacea-prone skin that cannot tolerate glycolic
- Darker skin tones (Fitzpatrick IV–VI) — where PIH risk from aggressive exfoliation is highest
- AHA beginners — as a gentler introduction to chemical exfoliation
- Acne-prone skin — particularly comedonal and inflammatory acne
- Post-inflammatory hyperpigmentation — where both exfoliation and tyrosinase inhibition are needed
How to Use It
Use at concentrations of 5–10% at a pH of 3.0–4.0. Apply to clean, dry skin 2–3 times per week, increasing frequency as tolerated.
