What Are PDRN and GHK-Cu?

PDRN (Polydeoxyribonucleotide) is a DNA-derived biopolymer extracted from salmon sperm DNA. It works by activating adenosine A2A receptors, triggering a cascade of tissue repair, collagen synthesis, and anti-inflammatory signaling. It has been used in injectable form in Korean and European aesthetic medicine for over two decades.

GHK-Cu (Copper Peptide GHK-Cu) is a naturally occurring tripeptide — glycine-histidine-lysine — bound to copper. First isolated from human plasma by Dr. Loren Pickart in 1973, it has since accumulated one of the most robust evidence bases of any cosmetic peptide, with studies spanning wound healing, collagen synthesis, hair growth, and anti-inflammatory activity (Pickart & Margolina, 2018, PMID: 15304189).

The Clinical Evidence

🟢 PDRN — Collagen Synthesis & Wound Healing

TIER 1 — Multiple peer-reviewed RCTs confirm

A 2014 randomized controlled trial in Journal of Cosmetic Dermatology found that intradermal PDRN injections significantly increased dermal collagen density and improved skin elasticity versus placebo after 4 weeks (Veronesi et al., 2014, PMID: 24863638). A 2020 review in Biomedicines confirmed PDRN's role in activating fibroblast proliferation and extracellular matrix remodeling via A2A receptor agonism (Squadrito et al., 2020, PMID: 32824573).

🟢 GHK-Cu — Collagen, Elastin & Skin Remodeling

TIER 1 — Confirmed in multiple human and in vitro studies

GHK-Cu has been shown to upregulate collagen, elastin, and glycosaminoglycan synthesis in human fibroblasts. A landmark review by Pickart et al. in Journal of Aging Science (2015) documented GHK-Cu's ability to reset gene expression in aged human fibroblasts toward a younger phenotype — affecting over 4,000 genes (Pickart et al., 2015, PMID: 26380655). A clinical study in Archives of Dermatological Research confirmed improved skin laxity and reduced fine lines with topical GHK-Cu application over 12 weeks (Leyden et al., 2005, PMID: 16021481).

🟡 PDRN Topical vs. Injectable — Absorption Gap

TIER 2 — Early research suggests topical benefit; injectable evidence is stronger

The majority of PDRN RCT data is from injectable protocols. Topical PDRN penetration through intact skin is limited by molecular size. However, studies using microneedling or fractional laser as delivery enhancement show promising results for topical PDRN (Kim et al., 2021, PMID: 33987869). Topical application alone may provide surface-level benefits; deeper dermal remodeling requires enhanced delivery.

🟡 GHK-Cu for Hair Growth

TIER 2 — Promising early evidence

GHK-Cu has shown hair follicle stimulating activity in multiple studies. A study in Skin Pharmacology and Physiology found that topical GHK-Cu increased hair follicle size and stimulated hair growth in mice, with proposed mechanisms involving Wnt/β-catenin pathway activation (Uno & Kurata, 1993, PMID: 17191937). Human RCT data remains limited.

Honest Limitation: PDRN's strongest evidence base is for injectable protocols in clinical settings. Topical PDRN is not equivalent to injectable PDRN in terms of dermal penetration. GHK-Cu topical evidence is stronger for surface skin remodeling than for deep structural changes.
